Indications for Use
Light Diagnostics SimulFluor® HSV1/2 Immunofluorescence Assay is a direct immunofluorescence test intended for the detection and identification of herpes simplex virus type 1 (HSV-1) or herpes simplex virus type 2 (HSV-2) following amplification in cell culture or by direct examination of clinical specimens prepared by cytocentrifugation. Specimens found to be negative on direct specimen examination should be tested by cell culture. For in vitro diagnostic use.
Device Story
SimulFluor® HSV 1/2 is an in vitro diagnostic immunofluorescence assay for detecting HSV-1 and HSV-2. Input: clinical specimens or cell culture samples. Process: reagent containing monoclonal antibodies specific to HSV-1 (glycoprotein C/capsid protein) and HSV-2 (glycoprotein G) tagged with distinct fluorescent labels is applied to slides. Unbound reagent is rinsed away. Output: visualization via fluorescence microscopy; HSV-1 appears apple-green, HSV-2 appears yellow-gold, and uninfected cells appear dull red (Evans blue counterstain). Used in clinical laboratories by trained technicians. Results assist clinicians in diagnosing HSV infections, enabling appropriate antiviral therapy. The single-reagent, simultaneous-detection format simplifies workflow compared to predicate methods requiring separate wells.
Clinical Evidence
Clinical evaluation conducted at two sites comparing SimulFluor® to predicate devices. Site 1 (n=191): Direct specimen sensitivity 89.5% (HSV-1) and 92.6% (HSV-2); specificity 100% for both. Site 2 (n=228): Compared in shell vials and 24-well plates; 100% agreement observed in 24-well culture plates. Overall performance demonstrated substantial equivalence to predicate devices.

Indications for Use
Indicated for the detection and identification of HSV-1 and HSV-2 in patients presenting with herpetic-like lesions. Specimens are obtained via direct clinical sampling or cell culture amplification. No specific age or gender contraindications noted.

Herpes simplex virus serological assays are devices that consist of antigens and antisera used in various serological tests to identify antibodies to herpes simplex virus in serum. Additionally, some of the assays consist of herpes simplex virus antisera conjugated with a fluorescent dye (immunofluorescent assays) used to identify herpes simplex virus directly from clinical specimens or tissue culture isolates derived from clinical specimens. The identification aids in the diagnosis of diseases caused by herpes simplex viruses and provides epidemiological information on these diseases. Herpes simplex viral infections range from common and mild lesions of the skin and mucous membranes to a severe form of encephalitis (inflammation of the brain). Neonatal herpes virus infections range from a mild infection to a severe generalized disease with a fatal outcome.
