Indications for Use
The TPO IgG kit is an Enzyme-Linked Immunosorbent Assay (ELISA) For the qualitative detection and quantitation of antibodies against the thyroid peroxidase antigen in serum as an aid in the diagnosis of thyroid autoimmune disease. The test can be performed either manually or in conjunction with the Mago Plus automated EIA processor.
Device Story
The anti-TPO IgG ELISA Kit is an in vitro diagnostic assay for detecting IgG antibodies to thyroid peroxidase in human serum. The device utilizes an enzyme-linked immunosorbent assay (ELISA) principle where TPO antigen is bound to microwells. Patient serum, calibrators, and controls are added; anti-TPO IgG antibodies bind to the antigen. After washing, a horseradish peroxidase-labeled anti-human IgG conjugate is added, followed by a substrate. The resulting colorimetric end product is measured spectrophotometrically at 450 nm. The absorbance is directly proportional to the concentration of IgG antibodies. The test is performed manually or using the Mago Plus automated EIA processor. Results assist clinicians in diagnosing thyroid autoimmune diseases. The device provides quantitative results in IU/ml.
Clinical Evidence
Clinical evidence includes retrospective testing of 255 patient sera compared to a commercial ELISA kit, showing 97.9% overall agreement. A second study of 207 characterized sera (normals, Grave's disease, Hashimoto's disease) demonstrated clinical specificity of 91.4% for normals and clinical sensitivity of 64.3% for Grave's and 75.0% for Hashimoto's disease. Precision studies (intra- and inter-assay) were conducted manually and on the Mago Plus processor, with CVs reported for various serum concentrations.
