Indications for Use

The Modified CEDIA DAU Phencyclidine Assay is a homogeneous enzyme immunoassay for the qualitative and semi-quantitative assay of phencyclidine in human urine. Measurements are used as an aid in the diagnosis and treatment of phencyclidine use or overdose.

Device Story

The CEDIA DAU PCP Assay is a homogeneous enzyme immunoassay using recombinant DNA technology to detect phencyclidine in human urine. The assay utilizes a bacterial enzyme, beta-galactosidase, engineered into two inactive fragments that reassociate to form an active enzyme. In the presence of the drug, the drug in the sample competes with drug conjugated to an inactive fragment for antibody binding sites; drug binding to the antibody leaves the inactive fragments free to reassociate and form active enzyme. The active enzyme cleaves a substrate, producing a color change measured spectrophotometrically. The absorbance change is proportional to the drug concentration in the sample. The assay is intended for clinical laboratory use to assist in diagnosing and treating PCP use or overdose.

Clinical Evidence

Bench testing only. Precision studies (N=126) showed total %CV ranging from 1.4% to 1.5% across concentration levels. Qualitative sensitivity at 25 ng/mL cutoff was 1.05 ng/mL. Accuracy compared to the predicate device showed 96.9% sensitivity and 100% specificity at the 25 ng/mL cutoff. Interference testing evaluated various substances (e.g., acetone, ascorbic acid, creatinine, ethanol) with less than 10% error observed.
