Indications for Use
The Epstein Barr Virus Viral Capsid Antigen (EBV-VCA) IgG ELISA Test is for the qualitative detection of IgG antibodies to EBV-VCA in serum as an aid in the clinical laboratory diagnosis of EBV infection in patients with clinical symptoms consistent with infectious mononucleosis (IM). The PANBIO EBV-VCA IgG ELISA should be used in conjunction with other EBV serologies.
Device Story
The EBV-VCA IgG ELISA Kit is an enzyme-linked immunosorbent assay used in clinical laboratories to detect IgG antibodies to Epstein-Barr virus viral capsid antigen (EBV-VCA) in human serum. The device utilizes microwells coated with EBV-VCA gp125 antigen derived from P3HR1 cell lines. Patient serum is added to the wells; if specific IgG antibodies are present, they bind to the immobilized antigen. After washing, peroxidase-conjugated anti-human IgG is added, followed by a TMB/H2O2 substrate system. The enzymatic reaction produces a blue color, which turns yellow upon the addition of an acid stop solution. The intensity of the color, measured via spectrophotometry, indicates the presence of EBV-VCA IgG antibodies. Healthcare providers use these results in conjunction with other EBV serological tests to assist in the diagnosis of EBV infection in patients with symptoms of infectious mononucleosis.
Clinical Evidence
Clinical performance was evaluated using 508 total serum samples across two study sites. Site 1 (retrospective, n=156) showed 69.2% sensitivity for acute cases, 94.1% for past infection, and 96.4% specificity. Site 2 (prospective, n=352) showed 92.9% sensitivity for acute cases, 89.3% for past infection, and 91.7% specificity. Analytical specificity was confirmed with a 29-specimen cross-reactivity panel (CMV, VZV, HSV-1, HSV-2, ANA, Rheumatoid Factor), showing 0% cross-reactivity. Reproducibility was assessed via ANOVA across three sites, with total CVs ranging from 4.7% to 14.1%.
